Efficient CAR-T Cell Characterization using Dry DURACloneAntibody Panels and Cytobank Analysis Software

CAR-T cell therapies are becoming a more common approach for treating hematological disorders. Flow cytometry is increasingly being used to comprehensively characterize CAR-T cell efficacy, as it can provide multiparametric data at single-cell resolution from starting material to final product, covering different CAR constructs, transfection methods and targets, enabling simultaneous monitoring of critical parameters and interrogating complex interrelated responses.


In this application note, we show how the characterization of CAR-T cells using preconfigured DURAClone and custom LUCID dry antibody panels helps laboratories overcome common challenges faced when using liquid antibody cocktails in flow cytometry experiments. These challenges include operator-dependent pipetting impact on accuracy and precision, and inconsistent reagent stability. To further streamline the workflow, acquired data is analyzed with Cytobank software using manual gating and advance machine learning-assisted tools to enumerate and visualize leukocyte cell subsets, T-cell maturation profiles and expression of CAR in samples during the different stages of the CAR-T cell development process from source material to the final product as depicted in the diagram below.
